Immunofluorescent assay of de novo vimentin expression in ovarian cancer tissues: surgical specimens vs paraffin-embedded tissue blocks

Abstract

The results of analysis of epithelial-mesenchymal transition in serous ovarian cancer were compared by the developed method of double immunofluorescent staining and flow cytometry of different tumor material types (surgical specimens vs paraffin-embedded tissue blocks). Estimation of de novo coexpression of mesenchymal protein vimentin in epithelial tumor cells expressing cytokeratins showed that the results were similar not only in quantitative terms, but also visually in the way of cell distribution on flow cytometry dot plots. The obtained results indicate prospects of the developed new technology for molecular diagnostics of tumor epithelial-mesenchymal transition into routine laboratory practice.
