Optimization of instrumental
parameters of the thermal lens detector for capillary electrophoresis
Abstract
The main instrumental parameters of the crossed-beam thermal
lens detector for capillary electrophoresis are considered. Optimum values of
the photodetector aperture size, distance between the capillary and the probe
and excitation beam waists, chopper frequency are calculated from the thermal
lens theory and confirmed experimentally. Under optimized conditions, the
separation of 2-, 3-, 4-nitrophenols is carried out. The limits of detection
(S/N=3) for nitrophenols are n´10–6 –
n´10–7
mol L–1. For these test substances the limits of detection
were improved by a factor of 30 compared to conventional UV-photometric
detection under the same conditions.
